The duo have won a combined total of 121 caps for Queensland, with coach Ewen McKenzie looking to utilise them in helping develop the club's rising stars.
"It's important to have the right balance of veterans and rookies in your squad and both Greg and Van bring a wealth of experience to the group while they are still playing at a very high level," said McKenzie.
"Next year we'll encounter a longer season again with an August finish and there will be more of a burden on the playing group. Having players with this sort of experience will help us see out a potentially more gruelling campaign."
Prop Holmes made his debut against the Blues in 2005 and was an integral member for the Reds as the Reds won the Super Rugby title.
"All the hard years were tough, but this year has made it all worthwhile and I'm happy to stay in Queensland," Holmes said.
"I've never wanted to leave and if Queensland is happy to keep me, I probably never will. Winning that title in something I never thought would happen and it has made it all the sweeter.
"We've got a really good squad here and everyone is happy. There is no reason we can't win it again and everyone is itching to get out there and go again next year."
Humphries first played for the Reds in 2000 and after a brief period with the Waratahs returned to Queensland for a second time in 2004.
"It's pretty amazing to win a Super Rugby title and you always hope to play in a team that achieves that sort of success. Now we've tasted it, every player is looking forward to next year," Humphries said.
"The Reds have generated a lot of interest in Rugby, not only in Queensland but around Australia, and it's exciting for us as a group to know people will be watching us. We're really looking forward to entertaining next year.
"When I went overseas in the middle of my career, playing 50 games is something I never thought I'd ever get, but it was amazing and I'm looking forward to getting a few more next year."
